China to conduct clinical trials of drugs effective in treating ulcerative colitis

IBD is a chronic inflammatory disease of the intestine, resulting from an excessive and prolonged immune response in the gastrointestinal mucosa due to a certain antigen (food/bacteria...) on a particularly sensitive genetic basis.

This is a disease with remission and relapse phases, which can last a lifetime. Interleukin-6 (IL-6) is thought to be a key mediator in the pathogenesis of IBD.

Current drugs are often ineffective in treating IBD, so a team led by researchers at China's Sun Yat-sen University has come up with a treatment strategy that controls symptoms and prevents relapses by blocking excessive immune responses.

Olamkicept is an improved soluble synthetic protein that can inhibit IL-6 signaling.

In the phase 2 clinical trials, the research team - with the participation of experts from 22 hospitals and research centers in East Asia - conducted a double-blind, randomized, placebo-controlled study.

A double-blind study is a study in which neither the researchers nor the participants know who is receiving which treatment during the trial.

During the study, which began in February 2018, researchers randomly assigned 91 patients with IBD, with an average age of 41, to three groups.

Patients received intravenous olamkicept at doses of 600 milligrams, 300 milligrams, or placebo, respectively, for 12 weeks.

Results showed that a 600-milligram dose of olamkicept showed a therapeutic effect in patients with moderate to severe IBD by significantly improving their clinical symptoms and biological markers.

Study leader Chen Minhu said the drug olamkicept was safe, well tolerated and could effectively reduce complications and mortality in patients with IBD.

The drug will enter phase 3 clinical trials this year.

The results of the study were published in the Journal of the American Medical Association (JAMA).
